Potential fully diluted market cap
$2,822.8M – $2,903.7MAward-overlap sensitivity range · historical share snapshot
June 30 capital snapshot, valued at the 2026-10-23 price ($24.05). 103,264,636 issued common shares + 12,628,958 outstanding options + 1,404,988 unexercised pre-funded warrants + 74,517 ESPP rights = 117,373,099 shares. The issuer groups 3,361,965 unvested RSUs and restricted shares together; excluding all or treating all as incremental gives the displayed overlap-sensitivity range. It is not a verified fully diluted point estimate or a bound on future issuance. All options are counted regardless of strike; no exercise-proceeds deduction. Excludes ungranted plan capacity and later share changes. Revalued on the 2026-10-23 regular-session price; share-component dates remain 2026-06-30. This weekly market update does not verify subsequent instrument changes or establish a current fully diluted capitalization.

| BEAM-302 Phase 1/2; pivotal expansion dosing underway [2] | Alpha-1 antitrypsin deficiency (AATD) SERPINA1 PiZ mutation: deficient functional AAT leaves lungs vulnerable; mutant protein accumulates in liver. | Liver-directed lipid nanoparticles deliver an adenine base editor to correct PiZ and restore functional M-AAT. |
| Risto-cel · BEAM-101 Phase 1/2 BEACON; adult/adolescent dosing complete [4][1] | Sickle cell disease with severe vaso-occlusive crises HbS polymerization drives red-cell sickling. Raising fetal hemoglobin (HbF) inhibits sickling. | Ex vivo editing of HBG1/2 promoters in autologous CD34+ cells prevents BCL11A binding; cells are reinfused after busulfan conditioning. |
| BEAM-304 IND cleared; Phase 1/2 start-up [6] | Phenylketonuria (PKU) PAH mutations impair phenylalanine metabolism; excess phenylalanine is neurotoxic. | Liver-targeted base editing, initially for R408W; mutation-specific editors are planned. No human efficacy disclosed in reviewed sources. |
| BEAM-301 Phase 1/2 dose exploration [7] | Glycogen storage disease type Ia (GSDIa) G6PC1 dysfunction impairs glucose release and causes fasting hypoglycemia. | Liver-directed base editing targets R83C to restore enzyme function. Initial clinical results were guided for 2026, not yet verified here. |
| BEAM-103 / ESCAPE BEAM-103 Phase 1 healthy-volunteer dosing complete [1] | Conditioning for hematopoietic stem-cell therapies CD117 marks hematopoietic stem cells; depletion creates space for replacement cells. | Anti-CD117 antibody intended to enable conditioning without genotoxic chemotherapy; clinical disease efficacy remains unproven. |
Clinical evidence already disclosed
- Population & design
- Open-label; 29 single-dose patients, cutoff June 24, 2026. Part A 60 mg: n=6; Part B 60 mg: n=5.
- Efficacy
- 60 mg: mean steady-state total AAT 14.4 µM (A) / 13.5 µM (B), versus baselines 5.0 / 4.7. Mutant Z-AAT fell 84%; corrected M-AAT comprised 93%.
- Safety
- Infusion reactions in 41%; one Part B patient had transient Grade 3 ALT/AST increases without bilirubin increase. Earlier repeat-dose cohort: one Grade 4 ALT event after the second dose.
- Interpretation
- Small, uncontrolled biomarker study; these results do not establish reduced emphysema, cirrhosis or mortality. Repeat-dose safety differs from single-dose results.
- Population & design
- 31 treated patients; cutoff August 6, 2025; follow-up 0.3–20.4 months. Open-label, single arm.
- Efficacy
- Mean HbF >60%, HbS <40%; no investigator-reported severe vaso-occlusive crises after engraftment. Anemia resolved after transfused blood cleared.
- Safety
- Transplant and busulfan risks remain. An earlier EHA report documented one death from respiratory failure four months after infusion, considered likely related to busulfan conditioning.
- Interpretation
- Unequal follow-up; not all 31 patients have long-term observation. Single-arm results do not prove superiority to other gene therapies.
Cash runway
Management guidance (August 4, 2026) assumes an additional $200m draw from the Sixth Street facility, alongside June cash and securities. The planned borrowing is not existing cash; this is not an independent burn-rate estimate. [1]
Latest verified financing
Latest verified priced equity offering: 16,151,686 common shares at $28.48 plus 1,404,988 pre-funded warrants at $28.47 with a $0.01 exercise price; approximately $500m gross. This is the issued price, excluding ATM sales and debt financing. [8]
